From: Interleukin-35 inhibited the production of histamine and pro-inflammatory cytokines through suppression MAPKs pathway in HMC-1 cells

IL-35 down-regulates the mRNA expression of IL-6 and IL-17 in HMC-1 stimulated by PMA and A23187. HMC-1 cells were treated with recombinant IL-35 (50 ng/ml and 100 ng/ml) for 6 h and stimulated with PMA (50 nM) plus A23187 (1 μM) for 8 h. After that, mRNA expression of IL-4, IL-6, IL-17, IFN-γ, and TNF-α in HMC-1 cells was measured by Real-time quantitative PCR. IL-35 inhibited the expression of IL-6 and IL-17 mRNA levels in HMC-1 cells. The mRNA expression of IL-4, IFN-γ, and TNF-α in simulated HMC-1 could not be inhibited by IL-35. All data are expressed as mean ± SD. P values are based on the One-way analysis of variance. n = 10
